Bigot, Laetitia.
The pacific oyster Crassostrea gigas exhibits an annual cycle of reproduction, which occurs in the gonadic area, a mix tissue composed of germinal cells and storage cells that regress during gametogenesis. Neuroendocrine regulations implicated in the physiological balance linking these tissues are not well known. Neuropeptide Y/F, which regulates energetic balance in vertebrates and invertebrates, binds G protein-coupled receptors; five related receptors have been identified in C. gigas. They display a differential pattern of expression depending on reproductive stage, and a higher expression in starved animals, compared to fed animals. By reverse endocrinology, no binding of NPY/F to these receptors could be demonstrated, but one of the receptors...

Characteristics of the particulate matter during the Mai '92 experiment. Effect on ecophysiological functions on mussel and oyster populations. ArchiMer
Prou, Jean; Soletchnik, Patrick; Heral, Maurice; Geairon, Philippe; Razet, Daniel; Bougrier, Serge.
In order to complete the knowledge of seasonal variability of ecophysiological parameters, in situ experiment was conducted in May 1992 at the sampling site "Le Chapus". As in the previous experiments (May '91, October '91 and February '92), the race-way technique was used to study ecophysiological behavior of oyster and mussel populations submitted to natural variability.

Cycle de reproduction naturelle de l'huître creuse Crassostrea gigas ArchiMer
Goulletquer, Philippe.
Since its introduction to French coasts in the 1970s, Crassostrea gigas completes its reproductive cycle—i.e. regular annual recruitment and establishment of natural populations—in only two sectors (Charentes coast and Arcachon Bay). In other areas of Pacific oyster production (Normandy, Brittany, Vendée, Mediterranean), complete gametogenesis may occur, resulting in spawning events and even recruitment in favourable temperatures. Temperature seems to be the main limiting factor for in situ reproduction. Growth and reproduction occur simultaneously. The study of the reproductive cycle of one-year old oysters showed that gametogenesis was asynchronous with high interannual variability and growth appeared to take precedence over reproduction. On the...

Development of a new diagnostic tool (Mini-Array) for ostreid Herpesvrus 1 (OsHV-1) detection in the Pacific oyster Crassostre gigas ArchiMer
Varsamos, S.; Pepin, Jean-francois; Sauvage, Christopher; Renault, Tristan.
OsHV-1 was the first herpesvirus to be identified in an invertebrate host and is associated to mortalities in the Pacific oyster (Crassostrea gigas) and other economically important bivalve species. Infections due to OsHV-1 are frequently reported and cause high mortality rates in larvae and spat of C. gigas. Detection of this pathogen is usually performed by PCR and/or in situ hybridization. This presentation will focus on the development of a new molecular diagnostic tool for OsHV-1 (and OsHV-1 variant 1) detection: the mini-array. This detection system, which allows multiple pathogens detection, combines viral DNA amplification by PCR and a specific hybridisation on a nylon membrane, revealed by a colorimetric reaction (DNA chip). Multiplex PCR involves...

Effects of temperature on herpes-like virus detection among hatchery-reared larval Pacific oyster Crassostrea gigas ArchiMer
Le Deuff, Rose-marie; Renault, Tristan; Gerard, Andre.
This paper describes effects of temperature on herpes-like virus detection in Pacific oyster Crassostrea gigas larvae held at different temperatures. Intranuclear, intracytoplasmic and extracellular viral particles were observed in velum and mantle connective tissues of oyster larvae reared at 25-26°C. In larvae held at 22-23OC, although nuclear lesions were observed, the presence of viral particles was not detected. Results were obtained for oyster larvae with 4 different parental origins. Herpesvirus infection was found in 3 of the 4 groups of oyster larvae held In the same conditions at the higher temperature.

Pien, Sebastien; Maheux, Franck; Gauquelin, Thibaut; Le Gendre, Romain.

Impact of intensive larval rearing practices in oyster hatcheries : selective processes and loss of genetic variability (slides) ArchiMer
Taris, Nicolas; Sauvage, Christopher; Boudry, Pierre.
Oysters, like many marine bivalves, are characterized by very high fecundity and low survival rate at early stages of life. Rearing conditions in hatcheries (i.e. density, feeding, temperature...) are different from those observed in the wild. Additionally, slow growing larvae are culled by sieving. As previous studies have shown that genetic variability exists for several early development traits, we aimed to assess if common hatchery rearing practices exert specific selective processes at the larval stage. In order to avoid possible bias and to increase the number of families studied, we used a mixed-family approach with subsequent parentage analysis using a recently developed set of three multiplexed microsatellite markers. We studied the variance in...

In vitro culture of mollusc hemocytes. Functional study of burst respiratory activity and analysis of interactions with protozoan and procaryotic pathogens ArchiMer
Boulo, Viviane; Hervio, Dominique; Morvan, Annie; Bachere, Evelyne; Mialhe, Eric.
The chief importance of hemocytes as immune effectors in molluscs has led to develop researches for in vitro culture. The chemiluminescence technique has been used to estimate the phagocytosis capacity of Crassostrea gigas hemocytes according to different culture media and times. Sea water was used as a basic medium. Additives, such as antibiotics or glucose and buffers, such as Tris or Hepes, were tested as weIl as artificial sea water. Hemocyte chemiluminescence activities were recorded at t=o in relation to the medium composition. The highest values were observed when hemocytes were kept in sea water without any modification. Whichever medium, chemiluminescence activity was substantially decreased when hemocytes were previously kept in vitro for a few...

Intensive rearing of juvenile oysters Crassostrea gigas in an upwelling system : optimization of biological production ArchiMer
Bacher, Cedric; Baud, Jean-pierre.
Seasonal growth of juvenile oysters (Crassostrea gigas) kept in an intensive upwelling system was studied from 1984 to 1986 in 60 to 90-day growth experiments. Saline ground water was used to produce Skeletonema costatum which was injected as food into the water supplying the upwelling system. Oyster density, water flow, phytoplankton concentration, temperature and frequency of food addition were controlled. Oyster growth was analysed with multidimensional contingency tables and correspondence analysis. The factors, in order of decreasing influence on growth, were temperature, food concentration and oyster density. A rearing strategy was deduced from the growth analysis for summer and winter. In summer, the density of 120,000 oysters/m2, the flow rate of...

His, Edouard; Tige, Gilbert; Rabouin, Marie-agnès.

Benabdelmouna, Abdellah; Grouhel, Stephanie; Cantin, Christian; Grizon, James; Seugnet, Jean-luc.
The monitoring of spat ploidy collected in the two main collection basins-Marennes Oléron and Arcachon-was realised through flow cytometry. Natural spat samples were collected on 3 sites in each basin and a total of 1 002 animals were analysed. Based on our samples, the results concluded that no polyploid, triploid or tetraploid animals were collected in the two investigated basins. These results comply with our provisions and do not bring any reason for a change in the practices used in triploid oyster culture. However, complementary studies on triploid residual gametogenesis, the nature of their gametes and that of their offsprings will have to be continued in order to fine-tune the estimates of the parameters on which the modeling scenarii of the...

Scanning the genome of the Pacific cupped oyster, Crassostrea gigas, for adaptative differentiation during geographic range expansion ArchiMer
Rohfritsch, Audrey; Bierne, Nicolas; Boudry, Pierre; Heurtebise, Serge; Lapegue, Sylvie.
Originating from the north eastern Asia, Crassostrea gigas has been introduced and translocated, mainly for aquaculture purpose, into several European countries (from Norway to Portugal and in the Mediterranean Sea) (1). Although highly variable, the invasiveness pattern of C. gigas has been demonstrated in several countries and therefore considered as a pest or a noxious species in those areas (2 and 3).Our project aims at identifying the characteristics of such a flourishing species: can its success be explained by chance and/or global warming only or does it exhibit a more important potential of adaptation than other species? Therefore we developed a population genomics approach, known as "genome scan". It corresponds to the study of numerous loci...

As the emergence of new vibriosis in the European Atlantic coasts has been associated with mass mortalities of molluscs cuasing important economic losses, the project obectives are to provide knowledge and operational tools to evaluate the impact of vibriosis on farmed japenese oyster Crassostrea gigas. Facing the diversity of Vibrio species associated to bivalve disease outbreaks, the difficulties of identification and the detemrination of their pathogenic abilities we propose a double approach to correlate phenotype/geneotype of a bacteria with risk of disease : 1) by genotyping we will evaluate if specific strains are associated with disease. 2) by evaluating likely virulence mechanisms implicated in pathogenesis, we will characterise the genes...

Razet, Daniel; Heral, Maurice; Prou, Jean; Legrand, Jacqueline; Sornin, Jean-marc.
The continuous obtention of current (speed and direction) turbidity and phytoplanktonic fluorescence was achieved for 9 successive days during the month of May 1988. Each two hours, during the day, the faeces and pseudofaeces productions of 7 oysters have been sampled individualy. The filtration rate were calculated in continuous. It appeared that the concentration in particular matter and also the planktonic fluorescence fluctuated in direct relation with the water depth, the origin of the water masses and the speed of the wind. During the tidal cycle the mean filtration rate was constant which induced a food consumption largely variable. The oysters were always above the limit of pseudofaeces production which regulated the ingested quantity. Their...
